چکیده

The procedure for the in vitro reconstitution of active 50 S ribosomal subunits from Escherichiu coli first appeared in 1974 [ 11. The same group later reported the kinetic study of the reconstitution steps and gave the estimated sedimentation values of the reconstitution intermediates in sucrose gradient in 1976 [2]. Due to the relative difficulty in isolating the intermediates, no physical studies of these intermediates have yet been reported. We have isolated the first intermediate RI,,(,), which is homogeneous and stable under low salt conditions. We report here the physical characteristics of the RI,o(l) particle as determined by hydrodynamic methods and compare our results with the intact 50 S subunits, which have been characterized under the same buffer conditions.

The importance of the Escherichia coli ribosomal protein L16 for the reconstitution of the peptidyl-tRNA hydrolysis activity of peptide chain termination.

The incubation of the 50 S ribosomal subunit of Escherichia coli with 1.5 M LiCl yields 1.5c core particles inactive in the peptidyl-tRNA hydrolysis activity of in vitro termination. The omission of L16 alone from reconstitutions of the proteins into the core results in inactive ribosomes. The single omission of a number of other proteins, in particular L7/L12, L10, L25, L27, and L15, gives rib...

Incorporation of dinitrophenyl protein L23 into totally reconstituted Escherichia coli 50 S ribosomal subunits and its localization at two sites by immune electron microscopy.

Escherichia coli ribosomal protein L23 was derivatized with [3H]2, 4-dinitrofluorobenzene both at the N terminus and at internal lysines. Dinitrophenyl-L23 (DNP-L23) was taken up into 50 S subunits from a reconstitution mixture containing rRNA and total 50 S protein depleted in L23. Unmodified L23 competed with DNP-L23 for uptake, indicating that each protein form bound in an identical or simil...

The RNA binding properties of "native" protein-protein complexes isolated from the Escherichia coli ribosome.

The assembly of the Escherichia coli ribosome is based on many specific interactions between the ribosomal components. Direct evidence for single proteinRNA interactions has been well established (reviewed in ref. [l] ). Various observations provide circumstantial support for the existence of protein-protein complexes in the ribosome. For example, cooperative protein binding effects have been o...

ESCHERICHIA COLI HEAT-LABILE TOXIN B SUBUNIT: CONSTRUCTION AND EVALUATION OF PLASMIDS PROVIDING CONTROLLED HIGH LEVEL PRODUCTION OF THE PROTEIN

With the plasmid DNA from a clinical isolate of enterotoxigenic Escherichia coli (ETEC) H 10407 as template, PCR-mediated cloning of the sequence encoding the heat-labile toxin B subunit (L T -B) has been carried out. Then this sequence was recloned into the pTrc 99A and pET23a expression vectors to give the pJasmids pTRCLTB and pETLTB, respectively. After induction, the former plasmid provides...

Expression of Recombinant Protein B Subunit Pili from Vibrio Cholera

Background & Aims: Vibrio cholerae is a gram-negative bacterial pathogen that causes cholera disease. Following ingestion by a host and entry into the upper intestine, V. cholera colonizes and begins to emit enterotoxin. One of the most pathogenic factors of Vibrio cholera is toxin-coregulated pili (TCP). ToxinCoregulated pili is as the primary factor requiered for the colonization and insisten...
